This is a full-time, fixed term position for 4 years on Crick Terms and Conditions of Employment.

The position of Postdoctoral Fellow in Cell & Developmental Biology has opened to work on a research project aimed at deciphering the coordination of tissue growth and patterning by signalling pathways. The post-holder will develop general methods to control the activity of various signalling pathways at high spatial and temporal resolution. Using these tools, he/she will identify the growth-relevant targets of these pathways thus setting the stage for an understanding of how patterning and growth are coordinated during development. The post-holder will join the research group of Jean-Paul Vincent at the Crick and report to him as line manager.

Objectives

The purpose of this postdoctoral post is to help develop a strong research programme on the role of signalling in coordinating growth and patterning. The role of the successful applicant will be inititially to develop cutting edge methods of controling signalling activity in transgenenic animals. This will be based on inducible dimerisation to mislocalise key components of signal transduction pathways. Two means of controling dimerisation, one based on light and the other on temperature, will be devised and evaluated. The optogenetic approach will take advantage of recent development as demonstrated in yeast and cultured cells. The temperature-based method will involve collaborating with physical biochemists and computational biologists. The most effective method will be used to reversibly inhibit the activity of three signalling pathways in Drosophila, those activated by Wingless, Dpp, and TOR. This will be used to assess the immediate early effects of these signalling pathways on growth, proliferation and patterning genes.The successful applicant will be using a wide range of techniques such as genome engineering, optogenetics, molecular biology, biochemistry, and live imaging.

Duties

Research (85%). The post-holder will conduct research of the highest quality on devising means of control protein activity and applying them to understand how key signaling pathways control growth and patterning in wing imaginal discs of Drosophila.The postholder’s main responsibility will be to advance his/her project. He/she will be also expected to train junior members of the lab in relevant techniques and to contribute actively to the intellectual life of the lab and the institute. When appropriate, the postholder will also contribute to collaborative work with other laboratories in the UK and further afield.

Manuscript preparation, peer reviewing and presentation of research findings (15%). Includes regular attendance and participation at scientific meetings with Jean-Paul Vincent and at other relevant meeting at the Francis Crick institute. Also includes writing manuscripts, presentation of work at national and international conferences and assisting Jean-Paul Vincent in reviewing manuscripts.

Key responsibilities:

1. The post-holder will be responsible for the progress of his/her research project. He/she will be pro-active and continually re-assess how the project develops. He/she will be expected to discuss long-term direction and general strategy with the group leader.

2. The post-holder will take part in advising other members of the group, such as PhD students, summer students, and new postdoctoral fellows by providing technical guidance.

3. The post-holder will provide scientific suggestions and other input at group meetings into the ongoing research of other members in the lab.

Working relationships:

The post-holder will discuss strategy, long term aims and data interpretation with the line manager, be Jean-Paul Vincent. He/she will also interact with other research staff, e.g. scientific officers, post-doctoral fellows, PhD students, and members of other labs at the Crick. In addition, the post-holder will participate in discussions with our collaborators in physcial chemistry and computational biology. Good working relationship with Crick support staff will also be expected.

Person Specification

The post holder should embody and demonstrate our core Crick values: Bold, Imaginative, Open, Dynamic and Collegiate, in addition to the following:

Essential:

• PhD in the area of cell and developmental biology
• Excellent knowledge of cell biology
• Good command of molecular biology and molecular genetics
• Experience with in vivo microscopy
• Track record as evidenced by publications or submitted manuscripts in refereed journals
• Experience in Drosophila genetics
