Job Description

SUMMARY: Work within a research laboratory with focus on tissue based analysis of resistance mechanism of immune checkpoint and targeted therapy in kidney cancer. Collaborate and assist in the development of new assays and improve existing laboratory methods. Successful candidate must be able to design novel projects, create hypothesis driven experiment, maintain excellent descriptions of experiments performed, standardize and validate new methods and be able to interpret results. Computer skills such as entering data obtained from experiments into graphic and statistics programs as well as the generation of figures are essentials. The employee should provide scientific expertise and organizational skills in getting involved in various research projects involving tissue and cell culture, small animal models and human subject research. Will be responsible for coordinating procedures with collaborators. Performing technical laboratory duties in accordance with general and/or specific plans. Read, write and edit manuscripts and reports, as well as help with grant writing.

SCOPE: To coordinate and conduct experiments and design research projects in the areas of cell and molecular biology, and immunology in animal and human tissues. Will keep abreast with literature of interest to laboratory, write manuscripts and help with grant writing. Will confer with collaborators in Genitourinary Medical Oncology and translational and Human Subject research regarding new procedures and modify them according to the needs of the projects. Record and evaluate data obtained in accordance with methods. The employee should be able to coordinate with the clinical researchers related to human subject data,

ESSENTIAL FUNCTIONS:

Planning and Execution of Experiments: The employee will conduct and carry out research experiments and is required to discuss goals of projects with principal investigator. Collaborate on detailed verbal and/or written instructions on how to conduct an experiment in order to achieve the goals of the project. The employee must be capable of proper operation and maintenance of all laboratory equipment. The successful candidate will exercise initiative and inventiveness in planning, executing and interpreting data obtained from experiments. Must be able to communicate results to principal investigators and collaborators. Must have the ability to work with the PIs troubleshoot various technical problems related to experimental design and execution. The position will require detailed, legible handwritten records of all experiments performed.

Specific Tasks: Sound working knowledge and trouble shooting skills in the most of the following techniques: Flow cytometry, RT-PCR, immunostaining (IHC) of tissue sections, gel electrophoresis of proteins and nucleic acids, western blot analysis, mammalian cell culture, cyropreservation of cells, and preparation and analysis of DNA, protein and small molecule, and CRISPR-cas9 gene editing method, exome and gene profiling data analyses, etc.

Media and reagent preparation: Must be able to measure optical density by spectrophotometer and calculate sample concentrations. Must have a basic knowledge of solution and buffer preparation in which ionic strength and solution pH are critical for the proper execution of experiments. Should be able to maintain equipment such as plate reader, fluorescence microscope, centrifuges, and pH meters. Must have knowledge of solubility, chemical characteristics, and uses related to new drug development. The successful candidate must be able to accurately calculate quantities of chemicals required to prepare biological buffers, media and experimental reagents such as investigational drugs. Must exercise proper caution when handling toxic, hazardous, flammable or potentially carcinogenic agents.

Tissue culture and handling of hazardous biological materials: Maintains and establishes tissue culture cell lines, which requires good sterile techniques and the ability to observe and identify cell changes under the microscope, determine cell counts on the hemocytometer or coulter counter, perform colony counts and determine cell viability. Prepares sterile media and solutions. Prepares cells for cryogenic storage and re-establishment by the use of small vials, dry ice and liquid nitrogen. Precaution against blood-borne pathogens must be taken whenever patient-derived cells are used in accordance with the institutional Universal Precautions Plan. Some protocols occasionally require weekend manipulations, and cultures must be maintained over holidays and vacations through coordination with other staff. Must demonstrate and practice laboratory safety precautions. Must manually handle, store and dispose of hazardous materials in procedures specified to meet MDACC safety guidelines. The position requires the maintenance and analysis of various viable (human) cell lines established from human tumors, immortalized human cell lines and primary human cancer cells. Tests may periodically be performed for mycoplasma contamination. Requires use of single and multichannel micro-pipetors.
