The Department of Medicine at Huddinge is comprised of seven units, three research centers and around 70 independent research groups. The Center for Infectious Medicine (CIM) is a research unit within the new research facilities at ANA Futura, KI Campus Flemingsberg that offers a dynamic and international research environment with a focus on translational studies of the human immune system and infection-immunity primarily in humans.

Your mission

In this project, the postdoctoral candidates will work with advanced immunological methods to study immunopathogenesis in human tuberculosis (TB). A major task will be to design and set up multicolour panels for flow cytometry, including both myeloid cells and T cells. The panels will be used for assessment of phenotypic and functional responses of cells from TB patient samples and in vitro Mycobacterium tuberculosis (Mtb)-infected cell cultures.

The postdoc training will be conducted in Assoc. Prof. Susanna Brighenti group. The research performed in the laboratory of Assoc. Prof. S. Brighenti involves studies on the pathogenesis of Mtb infection, the causative agent of tuberculosis (TB). The group has a focus to study immune responses in human cells obtained from TB patients or in vitro cultures using advanced cellular and molecular techniques. The long-term goal of the group is to improve our understanding of how mycobacteria can manipulate the human immune system and use this information to develop new host-directed immunotherapies that could support conventional anti-TB treatment with antibiotics aiming to enhance patient recovery.

The postdoc candidate is expected to establish Mtb infection models using both human cells and avirulent or virulent strains in vitro. Cryopreserved cell samples from TB patients will also be used. Majority of the work will be conducted in a biosafety level 3 (BSL-3) laboratory. The postdoctoral fellow will be required to execute a variety of techniques that include bacterial growth assays, live-cell imaging, mass and flow cytometry, quantitative PCR, cell sorting and RNA sequencing, and multiplex assays for cytokine/chemokine analyses. The fellow will also perform a variety of cell culture and in vitro functional assays, and extensively analyse high-dimensional data.

Your profile

Qualified to be employed as a postdoctor is one who has obtained a doctorate or has equivalent scientific competence. Applicants who have not completed a doctorate at the end of the application period may also apply, provided that all requirements for a completed degree are met before the (intended) date of employment. This must be substantiated by the applicant's main supervisor, director or equivalent.

Applicants should have a strong background in TB research and human immunology. Previous experience from work in a BSL-3 laboratory is mandatory. Applicants with knowhow in handling both clinical materials and in vitro Mtb infection models will receive priority. Competence and previous experience in cell sorting and different RNA assays are also considered a mandatory qualification. Furthermore, practical and theoretical knowledge in multi- colour flow cytometry including analyses with FlowJo and unsupervised analyses such as UMAP is a requirement. Clear and concise communication and written skills in English are mandatory. The successful candidate will work in close collaboration with the other group members and is expected to have high social and team building skills. The postdoctoral fellow is also expected to organize, plan, execute, and analyse experiments in an independent manner and contribute scientifically to CIM and the Brighenti group.
