实验室介绍

王宇涛研究员本科毕业于兰州大学,博士毕业于北京大学,师从蒋争凡教授,博士期间研究工作揭示了Caspase家族成员在天然免疫稳态和维持细胞凋亡免疫沉默中的调控作用;之后在哈佛医学院Christophe Benoist和Diane Mathis实验室完成博士后训练,运用体内高通量Rainbow-CRISPR筛选系统研究核受体家族在免疫细胞分化与细胞命运调控中的作用。王宇涛研究员将于2025年6月正式加入首都医学科学创新中心。

主要研究方向

1.核受体在病毒诱导的代谢重编程和天然免疫应答中的调控机制。

2.核受体在肿瘤进展与抗肿瘤免疫调控中的作用及其治疗潜力。

3.核受体介导免疫应答性别差异的分子机制研究。

4.基于合成生物学的转录因子活性检测和精准治疗策略开发。

主要成果与贡献

1. 建立体内高通量“Rainbow-CRISPR”筛选系统,系统解析核受体家族成员在免疫细胞分化和稳态维持中的作用,揭示视黄酸受体通过转录调控和炎症小体活化双重机制调控GATA6⁺腹腔巨噬细胞命运(Immunity,2024,一作)。

2. 揭示TRAF6介导的ASK1 Lys6-连接的多聚泛素化修饰在肝脏炎症和纤维化的关键作用(Hepatology,2019,一作);发现NLK激酶是抑制肝糖异生的关键分子,揭示血糖稳态调控的新机制(Cell Metabolism,2021,并列一作)。

3. 阐明Caspase家族成员在抗病毒天然免疫稳态和维持细胞凋亡免疫沉默中的调控作用(Immunity,2017,一作;Molecular Cell,2019,并列一作)。

免疫方向博士后

一、岗位职责

1.参与课题组科研工作,独立承担相应科研课题;

2.协助课题组长指导研究生实验;撰写基金和申报课题等相关工作。

二、岗位要求

1.近三年已经或即将获得免疫学、肿瘤学、细胞分子生物学或其他生物医学领域博士学位,具有代谢调控或者抗病毒天然免疫信号转导研究背景者优先;

2.具有扎实的分子和细胞生物学背景,如细胞培养、基因编辑、流式分析等技术,在小鼠模型实验或者生物信息学(多组学分析)方面具有较强的专业技能优先;

3.以第一作者身份在领域内主流期刊上发表过或即将发表研究论文,能够独立开展研究工作,具备高度的科研热情和严谨的科研态度;

4.责任心强,具有良好的团队合作精神,具备基本的英文沟通与学术写作能力。

生物信息学方向博士后

一、岗位职责

1.参与课题组科研工作,独立或合作承担相应科研课题;

2.协助课题组长指导研究生,撰写基金和申报课题等相关工作。

二、岗位要求

1.近三年已经或即将获得生物信息学、计算生物或系统生物学等相关专业博士学位,具有表观遗传组学(ChIP-Seq,ATAC-Seq,RNA-seq等)及单细胞测序数据分析,开发复杂生物信息算法或流程的经验者优先;

2.熟练掌握C++、python或R语言,熟悉Linux操作系统,能够高效处理与分析大规模生物数据;

3.工作认真负责,有良好的沟通能力和团队合作精神;

4.有良好的英语表达和论文写作能力。

科研助理

一、岗位职责

1.参与课题组科研工作,承担相应科研任务;

2.负责实验室经费管理,耗材订购等工作。

二、岗位要求

1.具有免疫学、肿瘤学、细胞分子生物学等相关专业的本科及以上学位;

2.掌握常规分子克隆、细胞培养的操作原理,有实验室研究工作经历优先;

3.有责任心和团队精神,热爱创新与挑战;

4.具备基本的英文沟通、写作能力。

Laboratory Introduction

Dr. Wang’s Laboratory at CIMR is now hiring postdoctoral fellows and research assistants! The lab is dedicated to uncovering the regulatory mechanisms underlying infection, inflammation, and tumor progression, with a particular focus on the nuclear receptor (NR) family in viral infections, cancer immunity, and sex-based differences in immune responses. By integrating small-molecule drug development and synthetic biology approaches, we aim to develop and advance innovative therapeutic strategies for human diseases.

Dr. Wang received his bachelor’s degree from Lanzhou University and his Ph.D. from Peking University, under the mentorship of Prof. Zhengfan Jiang, where he studied the regulatory mechanisms of the Caspase family in maintaining antiviral innate immune homeostasis and preserving immunological silence during apoptosis. He then completed his postdoctoral training at Harvard Medical School in the laboratory of Drs. Christophe Benoist and Diane Mathis, where he employed an in vivo Rainbow-CRISPR screening system to investigate the roles of nuclear receptors in immune cell differentiation and cell fate decisions. Dr. Wang will officially join the CIMR as an Assistant Investigator in June 2025.

Major Research Projects

a. Exploring the functions of nuclear receptors in viral infection-induced metabolic reprogramming and innate immune responses.

b. Dissecting the functions of nuclear receptors in tumor progression and immune regulation.

c. Uncovering the roles of nuclear receptors in sex-based differences in immune responses.

d. Targeting disease-specific transcriptional activity for precision therapy using synthetic biology approaches.

Major Contributions

a. Developed an innovative in vivo Rainbow-CRISPR screening platform and systematically investigated the nuclear receptor family in the differentiation and homeostatic control of 28 immune cell types, revealing that retinoic acid receptors regulate the fate of GATA6+ large peritoneal macrophages via transcriptional control of cell differentiation and inflammasome-mediated control of cell death (Immunity, 2024, first author).

b. Discovered a novel TRAF6-mediated Lys6-linked polyubiquitination of ASK1, which plays a pivotal role in of hepatic inflammation and fibrosis (Hepatology, 2019,first author); and identified NLK as a key suppressor of hepatic glucose production by inhibiting gluconeogenesis (Cell Metabolism, 2021, first author).

c. Identified the critical roles of the Caspase family in maintaining antiviral innate immune homeostasis and preserving immunological silence during apoptosis (Immunity, 2017, first author; Molecular Cell, 2019, co-first author).

Postdoctoral Research Fellow

– Immunology

Main Responsibilities

a. Lead independent research projects and collaborate with lab members.

b. Assist the PI in mentoring graduate students and preparing grant proposals and manuscripts.

Qualifications

a. Ph.D. in immunology, oncology, molecular / cellular biology, or a related biomedical field (obtained within the past three years or expected soon).

b. Solid background in molecular and cellular biology, including cell culture, gene editing, and flow cytometry. Experience with mouse immune-mediated disease models or bioinformatics (e.g., multi-omics analysis) is preferred.

c. Proven track record as first author in high-impact journals or manuscripts under revision; capable of conducting independent research with strong scientific enthusiasm and rigor.

d. Strong responsibility, excellent teamwork and communication skills, and proficient in English communication and scientific writing.

Postdoctoral Research Fellow

– Bioinformatics

Main Responsibilities

a. Lead independent research projects and collaborate with other team members.

b. Assist the PI in mentoring graduate students and preparing grant proposals and manuscripts.

Qualifications

a. Ph.D. in bioinformatics, computational biology, systems biology, or a related field (obtained within the past three years or expected soon); experience in epigenomic data analysis (e.g., ChIP-Seq, ATAC-Seq, RNA-Seq), single-cell sequencing, or development of bioinformatics pipelines/algorithms is highly preferred.

b. Proficient in C++, Python, R, and Linux; capable of processing and analyzing large-scale biological datasets.

c. Highly responsible, self-motivated, with excellent communication skills and a collaborative mindset.

d. Proficient in English communication and scientific writing.

Research Assistant

Main Responsibilities

a. Support research projects independently or in collaboration with lab members.

b. Assist with daily lab operations, including the management of supplies and ordering of reagents.

Qualifications

a. Bachelor’s degree or above in immunology, oncology, molecular and cellular biology, or related fields.

b. Prior research experience in molecular biology and cell culture is preferred.

c. Self-motivated researcher with strong sense of responsibility, good communication skills, and teamwork ability.

d. Basic proficiency in English communication and scientific writing.
